ارزیابی تنوع ژنتیکی نمونه‌های Beta vulgaris subsp. maritima با استفاده از نشانگرهای مولکولی هسته و سیتوپلاسم

چکیده

چغندر ماریتیما (Beta vulgaris ssp. maritima) نقش مهمی در برنامه­های به­نژادی چغندرقند دارد و تحقیق در مورد تنوع ژنتیکی آن اهمیت زیادی دارد. تعداد 13 نمونه چغندر ماریتیما شامل 10 نمونه ایرانی و سه نمونه اروپایی در این تحقیق ارزیابی شدند. دو نوع نشانگر مولکولی ISSR و ماهوارک (minisatellite) برای بررسی ژنوم هسته و میتوکندری مورد استفاده قرار گرفت. تعداد 176 مکان با 13 آغازگر شناسایی شد که 147 مورد (84/27 درصد) چندشکلی نشان دادند. نتایج مشخص نمود که در بین نمونه­ها ارزش محتوای اطلاعات چند شکلی (PIC) به میزان 32درصد است.  آغازگرهای (AC)8YT و (GA) 8YC بیشترین میزان ارزش PIC (38%) را داشتند. آغازگر (CT)8RG با بیشترین شاخص نشانگر (MI) می­تواند برای تجزیه­های بیشتر مورد استفاده قرار گیرد. بر اساس نتایج تجزیه­های UPGMA و ANOVA همه نمونه­ها به دو گروه اصلی تقسیم شدند. تجزیه به مؤلفه­های اصلی (PCoA) با دندروگرام تنوع ژنتیکی نشان داد که نمونه­های ایرانی و اروپایی از یکدیگر متفاوت هستند. برای مطالعه تنوع سیتوپلاسمی، از چهار نشانگر ماهوارک استفاده شد. نمونه­ها به چهار گروه میتوکندریایی بر اساس تنوع آغازگرها تقسیم شدند. با توجه به نتایج توالی‌یابی برخی نمونه­های ایرانی هتروپلاسمی بودند.
