تعیین تنوع ژنتیکی و بیماریزایی جدایههای Fusarium oxysporum از چغندرقند در ایران
تنوع ژنتیکی و بیماریزایی 13 جدایه قارچ Fusarium oxysporumکه طی سالهای 1387-1378 از مزارع چغندرقند شش استان کشور ایران جمعآوری شده بود، مورد بررسی قرار گرفت. تنوع بیماریزایی آنها با استفاده از روش غوطه ور سازی ریشه گیاهچههای چهارهفتهای در سوسپانسیون اسپور در لوله آزمایش انجام شد. بر این اساس جدایهها به دو گروه بیماریزای قوی و ضعیف دستهبندی شدند. تنوع ژنتیکی جدایه ها با استفاده از ۱۵ آغازگر تصادفی و به روش RAPD ارزیابی شدند. بر این اساس چند شکلی قابل ملاحظه ای بین جدایههای مورد بررسی مشاهده شد. گروه بندی خوشهای جدایه ها با استفاده از UPGMA به کمک نرمافزار MVSP، جدایهها را به پنج گروه طبقه بندی کرد. رابطه مشخصی بین تنوع ژنتیکی بر اساس RAPD-PCR با بیماریزایی جدایهها یافت نشد، اما مجزا شدن برخی از جدایه ها از سایرین با مناطق جغرافیایی آنها در ارتباط بود. DNA ژنومی جدایهها با کمک آغازگرهای ITS1 و ITS4 تکثیر شد. محصول تکثیر شده از DNA جدایههای مختلف ، با آنزیمهای برشگر TaqI ,HaeIII ,EcoRI هضم گردید. آنالیز ITS-RFLP جدایه ها الگوی باندی مشابهی را نشان داد.

روند توسعه بیماری سفیدک پودری در مزارع چغندر قند در استان اصفهان
کشت چغندرقند در استان اصفهان از اهمیت خاصی برخوردار است. یکی از عوامل محدودکننده این محصول استراتژیک بیماری سفیدک پودری میباشد. جهت بررسی وضعیت آلودگی مزارع چغندرقند به بیماری سفیدک پودری در این استان به نمونهبرداری از نقاط مهم چغندرکاری شامل اصفهان، سمیرم، فریدن و کمشچه در دو سال 1388 و 1389 اقدام گردید. برای تعیین شدت آلودگی، در هر منطقه ده مزرعه مشخص گردیده و در شش مرحله مورد ارزیابی قرار داده شدند. شدت بیماری بر اساس صفر تا 100 اندازهگیری شد. نتایج حاصل از بررسی سفیدک پودری در مزارع چغندرقند در چهار منطقهی مورد مطالعه، نشان داد که در مجموع، میانگین شدت بیماری در مزارع مورد مطالعه در هر منطقه متفاوت و با به طور معنیدار از یکدیگر متمایز بودند. بیشترین شدت آلودگی در منطقه اصفهان و سمیرم به ترتیب 30/50 و31/46 درصد و کمترین شدت بیماری در دو منطقه کمشچه و فریدن به ترتیب 94/18 و44/14 درصد محاسبه شد. بیماری در مناطق اصفهان، فریدن، سمیرم و کمشچه از مرداد ماه شروع میشود. در اصفهان از اواسط شهریور ماه تا اواسط مهر ماه، در فریدن شهریور ماه، در سمیرم اوایل شهریور ماه و کمشچه در مهرماه به اوج خود میرسد. شدت بیماری در اصفهان و کمشچه تا آبان ماه و در فریدن و سمیرم تا اواخر مهر ماه تدریجاً ادامه داشت. فرم جنسی عامل بیماری در اواخر فصل به صورت نقاط سیاه رنگ (کلیستوتسیوم) مشاهده گردید. مطالعات میکروسکوپی تولید آسکوسپورها را در منطقه تأیید نمود.

بررسی الگوی توزیع و تغییرات جمعیت زنجرکهای ناقل بیماری کرلیتاپ چغندرقند در استان همدان
یکی از بیماریهای مهم چغندرقند کرلیتاپ میباشد. در این تحقیق الگوی توزیع فضائی و روند تغییرات جمعیت زنجرکهای (Baker) Circulifer haematoceps و C. tenellus ناقلین ویروس کرلیتاپ چغندرقند به مدت سه سال (1386 لغایت 1388) در مزارع چغندرقند همدان مورد مطالعه قرار گرفت. بررسیها در مراحل اولیه رشد گیاه با توجه به تاریخ کاشت مزارع به مدت نه هفته و به فاصله حدود هفت تا 10 روز از هم انجام شد. برای این کار، چهار مزرعهی پنج هکتاری با فاصله تقریبی یک کیلومتر از هم در شهرستان اسدآباد استان همدان انتخاب شدند. هر بار تور زدن به عنوان یک واحد نمونهبرداری تعیین شد. با استفاده از دادههای بهدست آمده فاکتور RV (معیار دقت نمونهبرداری) محاسبه شد که برای هر سال به ترتیب برابر 2/12، 52/6 و 65/16 درصد بود. با توجه به اینکه زنجرک گونه haematoceps C. تنها ناقل مشاهده شده در منطقه مورد بررسی بود، جهت تعیین الگوی توزیع فضایی جمعیت این زنجرک از روش نسبت واریانس به میانگین استفاده شد. مقدار عددی Z برای طول سال و مجموع سه سال بزرگتر از عدد 96/1 بود که نشان دهنده توزیع فضائی تجمعی ناقل میباشد. نتایج روند تغییرات و نسبت جمعیت زنجرک ناقل haematoceps C. به کل جمعیت سایر گونههای زنجرک در سالهای 1386، 1387 و 1388 به ترتیب برابر 68/5، 97/1 و 43/2 درصد بود. میانگین درصد آلودگی بوتهها به بیماری کرلیتاپ در این سالها به ترتیب برابر 2/5، 6/8 و 5 درصد بود. در سال 1386 با وجود بالا بودن تعداد و نسبت جمعیت زنجرک ناقل به کل جمعیت سایر گونهها، میانگین درصد آلودگی بوتهها به بیماری کرلیتاپ کمتر بود. این امر ناشی از تاخیر در تاریخ کاشت است. در حالی که در سال 1387 در نتیجه رشد کندتر بوتهها در ابتدای فصل رشد به دلیل کاشت زود هنگام، زمان بیشتری در اختیار ناقل قرار گرفته است. در این شرایط با وجود پایین بودن تعداد و نسبت جمعیت زنجرک ناقل به کل جمعیت سایر گونهها، میانگین درصد آلودگی بوتهها (6/8 درصد) بالاترین میزان آلودگی نسبت به دو سال دیگر را نشان داد. همچنین نتیجهگیری شد که بین فراوانی جمعیت زنجرک ناقل در مزرعه با فراوانی آلودگی بوتهها به ویروس کرلیتاپ در طول فصل رشد رابطه خطی مثبت و معنیدار وجود دارد.

این آزمایش با هدف تعیین تبخیر-تعرق گیاه چغندرقند و برآورد ضریب گیاهی (kc) آن برای دوره رشد با استفاده از لایسیمتر زهکشدار به روش بیلان آبی و روشهای تجربی به مدت سه سال در شهرکرد اجراء شد. پس از کاشت بذر چغندرقند در داخل و خارج لایسیمتر، با اندازهگیری عوامل معادلهی بیلان آبی، تبخیر-تعرق گیاه در دوره هفتگی و ماهانه محاسبه گردید. نتایج نشان داد که مجموع تبخیر-تعرق گیاه چغندرقند در فصل رشد برابر6/1016 میلیمتر شد که میزان آب زهکش برابر با 9/73 میلیمتر و تغییرات رطوبتی خاک برابر با 1/66 میلیمتر بود. مقدار تبخیر از تشت کلاس A در فصل رویش چغندرقند برابر 5/1364 میلیمتر شد. تبخیر-تعرق گیاه مرجع از طریق لایسیمتر زهکشدار اندازهگیری و با استفاده از روشهای تجربی برآورد گردید، نتایج نشان داد که میزان تبخیر-تعرق گیاه مرجع از لایسیمتر در دوره رشد گیاه چغندرقند برابر با 03/1123میلیمتر بود و از بین روشهای تجربی، روش بلینی–کریدل فائو 24 و پن من مانتیث فائو 56 از دقت بیشتری در برآورد برخوردار بودند. ضریب گیاهی چغندرقند یا (kc) در مراحل رشد ابتدایی، توسعه گیاه، میانی و نهایی به ترتیب برابر با 72/0، 81/0، 04/1 و 70/0 شد و متوسط آن در کل دوره رشد گیاه چغندرقند برابر با89/0 بود. متوسط ضریب تشت تبخیر یاkpدر طول دوره رویش گیاه چغندرقند برابر با 83/0 شد و متوسط ضریب (Kc.p) در فصل رشد گیاه چغندرقند برابر با 73/0 به دست آمد. به عبارتی نیاز آبی چغندرقند برابر با 73/0 تبخیر از تشت تبخیر است و با داشتن میزان تبخیر از تشت با دقت قابل قبولی میتوان نیاز آبی گیاه را تخمین زد. از طرفی کارایی مصرف آب برای محصول ریشه چغندرقند برابر با 14/5 کیلوگرم بر مترمکعب و برای شکر سفید برابر با 753/0 کیلوگرم قند بر مترمکعب آب مصرفی برآورد گردید.

اثر تنش شوری بر اجزای فتوسنتزی چغندرقند در شرایط گلخانه و مزرعه
اندازهگیری میزان کلروفیل و فلورسانس کلروفیل دارای پتانسیل لازم برای آنالیز کارایی فتوسنتز گیاهان در برابر تنشهای محیطی به ویژه تنش شوری بوده و کاربرد آسان آن، مطالعه وضعیت تنشها را تسهیل مینماید. به منظور بررسی واکنش صفات مربوط به دستگاه فتوسنتزی نسبت به تنش شوری در مراحل مختلف رشد ژنوتیپهای چغندرقند، دو آزمایش مجزا طراحی و اجرا شد. شش ژنوتیپ چغندرقند تحت دو تیمار اولی بدون تنش (شاهد) و دومی شوری با هدایت الکتریکی 16 دسی زیمنس بر متر در گلخانه و مزرعه ارزیابی شدند. نمونهگیری در گلخانه طی مراحل چهار برگی و استقرار (هشت برگی) و در مزرعه طی مراحل رشد برگی (16 برگی) و مرحله رسیدگی فیزیولوژیک (40 برگی) انجام شد. در مراحل نمونهگیری کارایی فتوسیستم II، متغیرهای تبخیر و تعرق، هدایت روزنهای، فتوسنتز، تنفس و مقادیر کلروفیل a و b اندازهگیری شد. بیشترین اثر شوری از نظر صفات فتوسنتزی در مراحل مختلف رشد در مرحله دوم رشد چغندرقند (8 تا 10 برگی یا استقرار) مشاهده گردید. همبستگی معنیداری بین میزان تعرق برگی، هدایت روزنهای و کل مقدار کلروفیل با عملکردهای ریشه و قند مشاهده شد. در تیمار شوری طی مرحله اول رشد با کاهش فلورسانس اولیه و عدم تاثیر روی فلورسانس حداکثر، کارایی فتوسیستم IIافزایش یافت اما در مرحله استقرار گیاه کاهش کلیه پارامترهای فلورسانس کلروفیل، باعث کاهش معنیدار کارایی فتوسیستم II گردید که این امر موجب آسیب به ساختار فتوسنتزی گیاه و کاهش مقدار کل کلروفیل و کلروفیل a و bشد. تحمل ژنوتیپ 7219 نسبت به شوری با کاهش تعرق و هدایت روزنهای همراه بود اما دو ژنوتیپ BP Karaj و 7233 p.29*MSC2 با کاهش تعرق و افزایش فلورسانس کلروفیل نسبت به شوری تحمل نشان دادند. ژنوتیپ 452 نسبت به شوری حساس بود و هیچ یک از مکانیزمهای تحمل به تنش شوری در آن مشاهده نشد. در نهایت مشخص گردید که علاوه بر این که ژنوتیپهای مختلف چغندرقند مکانیزم متفاوت فیزیولوژیکی برای تحمل به تنش شوری دارند، مراحل مختلف رشد نیز در بروز واکنش فیزیولوژیک تأثیرگذار است. بنابراین در غربال ژنوتیپها مکانیزمهای فیزیولوژیکی تحمل به تنش در مراحل مختلف رشد نیز حائز اهمیت میباشد.

تأثیر میزان رطوبت بر برخی خواص فیزیکی بذر چغندرقند
در این تحقیق برخی خواص فیزیکی بذر دو رقم چغندرقند به نامهای "شیرین" و گدوک (436) به عنوان تابعی از محتوای رطوبت بذر مورد بررسی قرار گرفت. این خواص عبارت از: طول، عرض، ضخامت، میانگینهای حسابی و هندسی ابعاد، ضریب کرویت، وزن هزار دانه، زاویه انباشتگی، سرعت حد، چگالی واقعی، چگالی ظاهری، تخلخل و ضریب اصطکاک ایستایی بودند. آزمایش به صورت فاکتوریل در قالب طرح کاملاً تصادفی شامل دو رقم بذر و چهار سطح رطوبتی بذر (به میزانهای 4/8، 8/9، 9/11 و 14 درصد) اجرا شد. نتایج نشان داد که با افزایش رطوبت، مقادیر طول، عرض و ضخامت بذر رقم "شیرین" به ترتیب 37/11، 61/15 و 82/8 درصد و بذر رقم گدوک (436) به ترتیب 5، 54/4، 69/6 درصد، افزایش یافتند. میانگینهای حسابی و هندسی ابعاد، ضریب کرویت، وزن هزار دانه، زاویه انباشتگی و سرعت حد نیز در هر دو رقم با افزایش رطوبت، افزایش یافتند. این در حالی بود که چگالی واقعی، چگالی ظاهری و تخلخل با افزایش رطوبت، به صورت خطی کاهش یافتند.ضریب اصطکاک بذور هر دو رقم، بر روی چهار سطح لاستیک، تخته سه لایه، فولاد گالوانیزه و آلومینیوم، با افزایش رطوبت افزایش یافتند. بیشترین ضریب اصطکاک برای بذر هر دو رقم بر روی سطح لاستیکی و کمترین آن بر روی سطح فولاد گالوانیزه به دست آمد. معادلات رگرسیونی مربوط به خواص فیزیکی اندازهگیری شده بر حسب محتوای رطوبت بذر تعیین و با استفاده از تجزیه واریانس مورد ارزیابی قرار گرفتند. نتایج نشان داد که معادلات حاصله میتوانند در سایر سطوح رطوبتی نیز برای پیش بینی خواص فیزیکی مورد بررسی در این تحقیق، به کار گرفته شوند.

بررسی تاثیر عناصر ریزمغذی بر برخی خصوصیات ریختشناختی مرتبط با کمیت و کیفیت بذر چغندرقند رقم شیرین
این تحقیق در دو سال زراعی 84-1383 جهت تعیین اثرات عناصر ریزمغذی بر برخی خصوصیات ریختشناختی مرتبط با کمیت و کیفیت بذر چغندرقند رقم شیرین در مزرعه ایستگاه تحقیقات کشاورزی اردبیل انجامشد. بر اساس نتایج تجزیه شیمیایی خاک مزرعه مورد آزمایش دارای واکنش 62/7 – 80/7، کربن آلی 53/0 – 49/0 درصد بوده و مقادیر نیتروژنکل، فسفر و پتاسیم قابل جذب آن نیز در حد مطلوب ارزیابی شدند. آزمایش به صورت طرح بلوکهای کامل تصادفی با سه تکرار اجرا گردید. تیمارهای آزمایش شامل هشت تیمار: (1) شاهد (بدون مصرف عنصر ریزمغذی)، (2) آهن Fe، (3) منگنز Mn، (4) بور B، (5) آهن و منگنز، (6) آهن و بور، (7) بور و منگنز و (8) آهن، بور و منگنز بودند. نتایج آزمایش نشان داد که تأثیر تیمارهای مختلف بر تعداد شاخههای فرعی، قطرشاخهاصلی، وزن خشکبرگ، وزن خشکشاخه و وزن خشککل بوته در سطح یک درصد معنیدار میباشد. از مقایسه میانگینها مشخص شد که بیشترین تعداد شاخهفرعی با مصرف عنصر آهن حاصل شده، بالاترین مقادیر وزن خشکبرگ، وزن خشکشاخه و وزن خشککل با مصرف آهن و نیز با مصرف توأم عناصر آهن و بر به دست آمده است. همچنین نتایج نشان داد که تأثیر عناصر ریزمغذی بر عملکرد بذر، درصد پوکی، درصد قوهنامیه، درصد بذر با سایز 5/4-5/3میلیمتر گرد (f) معنیدار بود و با مصرف توأم آهن و بر بیشترین عملکرد بذر در هکتار با کمترین درصد پوکی و بالاترین قوه نامیه حاصل گردید.

بررسی تناسب محیطی شهرستان تربت حیدریه برای کشت چغندرقند با استفاده از سامانه اطلاعات جغرافیایی
هدف اصلی این مطالعه، تعیین نواحی مستعد کشت چغندرقند با توجه به برخی متغیرهای اصلی مؤثر در آن میباشد. دادههای اقلیمی موردنیاز، از اداره کل هواشناسی خراسانرضوی که در ایستگاههای منطقه جمعآوری شده بود برای دوره 1388-1377 اخذ شد. جهت تهیه نقشه سطوح ارتفاعی، شیب، جهت شیب و TIN از نقشههای توپوگرافی سازمان نقشهبرداری کشور با مقیاس 1:250000 استفاده گردید. نقشه پوشش گیاهی و کاربری اراضی از نقشههای قابلیت اراضی در مقیاس 1:250000 مؤسسه تحقیقات خاک و آب کشور استخراج گردید. از سامانه اطلاعات جغرافیایی به منظور رقومیسازی و تهیه نقشهها بهرهبرداری شد. پس از تشکیل پایگاه اطلاعات فضایی منطقه، با استفاده از نرمافزار ARCGIS اطلاعات توصیفی نقشهها به آنها اضافه شد. سپس با توجه به اقلیم موردنیاز محصول چغندرقند و با استناد به نظر کارشناسان بر اساس یک مدل تصمیمگیری سلسله مراتبی، وزن پارامتر و اهمیت هریک از لایهها با استفاده از نرمافزارEXPERT CHOICE تعیین گردید. در نهایت با عملیات همپوشانی وزنی در محیط سیستم اطلاعات جغرافیایی(GIS) پهنهبندی عرصههای مستعد کشت چغندرقند در منطقه انجام گردید. در بین معیارهای تصمیمگیری نیازآبی و بارش مؤثر مهمترین عوامل تعیین گردیدند. نتایج حاصل از نقشه نهایی پهنهبندی نشان داد که61/59 درصد مساحت شهرستان معادل 82/3745 کیلومتر مربع از قابلیت خیلی خوبی برای کشت چغندرقند برخوردار میباشد و حدود 49/24 درصد از مساحت شهرستان نیز فاقد استعداد زراعی برای کشت این محصول میباشد. مناطق دشت رخ و بخش کدکن در شمال شهرستان تربت حیدریه به عنوان مستعدترین مناطق و پس از آن دشت مرکزی به عنوان منطقه مناسب مشخص شدند.
